English name: T7 RNA Polymerase

This enzyme is derived from E. coli and expressed as the bacteriophage T7 RNA Polymerase gene. It is a highly specific DNA dependent 5 "→ 3" RNA polymerase that recognizes the T7 promoter sequence. T7 RNA Polymerase can catalyze the incorporation of NTP downstream of single or double stranded DNA T7 promoter, synthesizing RNA complementary to the template DNA downstream of T7 promoter.

Features: T7 RNA Polymerase can recognize modified NTPs, such as biotin labeled, d-i-g-o-x-i-n labeled, and fluorescein labeled NTPs, and can be used for the synthesis of various labeled RNAs. At the same time, it has high specificity for the T7 promoter.

Activity definition: The amount of enzyme required to catalyze the incorporation of 1 nmol AMP into polynucleotides within 60 minutes at 37 ºC is defined as 1 active unit.

Activity detection conditions: 40mM Tris HCl (pH 8.0), 6mM MgCl2, 10mM DTT, 2mM spermidine, 0.5mM NTP, 0.6MBq/ml [3H] - ATP, 20 μ g/ml plasma DNA containing the specific T7 RNA polymerase promoter sequence.

Purity: No DNA endonucleases and exonucleases, no RNA enzymes.

Enzyme storage solution: 50mM Tris HCl (pH 8.0), 150mM NaCl, 5mM DTT, 0.1mg/ml BSA, 0.5mM Elugent, 50% glycol.

Translation Buffer (5X): 200mM Tris HCl (pH 7.9 at 25 ºC), 30mM MgCl2, 50mM DTT, 50mM NaCl, 10mM spermidine.

Deactivation or inhibition: Heating at 70 ºC for 10 minutes can inactivate T7 RNA Polymerase. Adding an appropriate amount of EDTA can also inactivate T7 RNA Polymerase. Chelating agents, sodium, potassium, or ammonium salts with concentrations greater than 150mM can significantly inhibit the activity of T7 RNA Polymerase.

Usage: Used for RNA synthesis. The synthesized RNA can be used as a hybridization probe, genomic DNA sequence analysis, RNase protection assay, antisense RNA synthesis, as an RNA template for in vitro translation, as a substrate for RNA splicing research, RNA secondary structure and RNA protein interaction, nucleic acid amplification analysis, siRNA, miRNA, and other small RNAs.
