Often used to degrade proteins and produce short peptides.

Protease K is an important serine protease secreted by Candida albicans. In 1974, Wolfgang Ebeling et al. isolated and purified protease K from the culture medium of Candida albicans using precipitation, ion exchange chromatography, and coagulation chromatography. Protease K belongs to the Bacillus subtilis family. It has high enzyme activity and wide substrate specificity, and is often used to degrade proteins and produce short peptides. It has the typical catalytic triad Asp39 His69 Ser224 characteristics unique to serine p-r-o-t-e-a-s-e-s, and two Ca2+binding sites around the active center increase its stability, allowing it to maintain high enzyme activity under a wider range of conditions.

Application:

1. Application in nucleic acid extraction and detection: Protease K can degrade histones tightly bound to nucleic acid, separate DNA from proteins, and purify DNA; Moreover, protease K can degrade RNA hydrolase activity (Rnase), inhibit the hydrolysis of template RNA by Rnase, and play an indispensable role in RNA extraction and detection.

2. Application in in situ hybridization: Protease K is commonly used to digest proteins around target DNA, facilitating probe penetration and improving detection sensitivity. It should be noted that high concentration of protease K or prolonged digestion time can damage the cell structure, leading to phenomena such as disappearance and detachment of the nucleus, affecting the binding of nucleic acids.

3. Application in non protein component extraction: Protease K can degrade protein impurities, separate target components from proteins from biological samples, improve product purity and production efficiency, especially for degrading harmful protein components such as endotoxins.
