DPPH method: The DPPH method was proposed in 1958 and is widely used for quantitative determination of biological samples, classification of substances, and antioxidant capacity of food. DPPH is a stable free radical in organic solvents, and its alcohol solution is purple in color. It needs to be stored in a low-temperature, dark place and has a single electron, so it can accept one electron or hydrogen ion. It has maximum absorption at a wavelength of 517nm. When free radical scavengers are present, the single electron of DPPH is captured, causing its color to become lighter. The absorbance value at the maximum light absorption wavelength decreases, and the degree of decrease is linear. The decrease in absorbance level indicates an increase in antioxidant activity, thus evaluating the antioxidant ability of the test sample. This antioxidant capacity is represented by inhibition rate, and the greater the inhibition rate, the stronger the antioxidant activity.
