Fibrinogen (Fg), also known as coagulation factor I, is derived from bovine plasma. It has a molecular weight of approximately 340KD and is composed of three different pairs of peptide chains: alpha, beta, and gamma. The alpha chain has a molecular weight of 63.5 kDa, beta chain has a molecular weight of 56 kDa, and gamma chain has a molecular weight of 47 kDa (containing approximately 4% carbohydrates). Peptide chains are connected by disulfide bonds. Under the action of thrombin, the alpha and beta chains release A and B peptides respectively, generating fibrin monomers. During this process, due to the release of acidic peptides, the negative charge is reduced, and the monomers are easily polymerized into fibrin polymers. But at this time, the monomers are connected by hydrogen bonds and hydrophobic bonds, and can still be dissolved in dilute acid and urea solutions. Furthermore, under the action of Ca2+and activated factor XIII, monomers are covalently linked to form stable insoluble fibrin clots, completing the coagulation process. Fibrinogen solution denatures above 50 ºC, so the heating temperature should not exceed 50 ºC; In addition, fibrinogen should generally be dissolved in physiological saline at 37 ºC, and the temperature should not be too low during dissolution. It is difficult to dissolve at 4 ºC and in saline free solutions.
